We presented a clear cause–mechanism–effect story: the need for better CNS delivery and reduced liver exposure, the JBC tag enabling BBB transcytosis and capsid de-targeting, and the resulting increase in CNS expression with lower off-target risk—positioning the approach as promising for rare neuro and neurodegenerative diseases.
